背景介绍

The nuclear lamina consists of a two-dimensional matrix of proteins located next to the inner nuclear membrane. The lamin family of proteins make up the matrix and are highly conserved in evolution. During mitosis, the lamina matrix is reversibly disassembled as the lamin proteins are phosphorylated. Lamin proteins are thought to be involved in nuclear stability, chromatin structure and gene expression. Vertebrate lamins consist of two types, A and B. Alternative splicing results in multiple transcript variants. Mutations in this gene lead to several diseases: Emery-Dreifuss muscular dystrophy, familial partial lipodystrophy, limb girdle muscular dystrophy, dilated cardiomyopathy, Charcot-Marie-Tooth disease, and Hutchinson-Gilford progeria syndrome. Western blot analysis of extracts (10ug) from 3 different cell lines by using anti-LMNA monoclonal antibody (1:1000).

HEK293T cells were transfected with the pCMV6-ENTRY control (Left lane) or pCMV6-ENTRY LMNA (Right lane) cDNA for 48 hrs and lysed. Equivalent amounts of cell lysates (5 ug per lane) were separated by SDS-PAGE and immunoblotted with anti-LMNA(Cat# BD-PE5384). Positive lysates (100ug) and (20ug) can be purchased separately from OriGene.

Immunofluorescent staining of Hela cells using anti-LMNA mouse monoclonal antibody (BD-PE5384, green, upper left; merged, upper right) or Isotype control ( merged, lower right). Cell nuclei were stained with DAPI (blue, lower left) (1:100).

Western blot analysis of extracts (35ug) from different cell line by using anti-LMNA monoclonal antibody (1:500).
